Effects of parathyroid hormone type 1 receptor on inflammatory factors and angiogenesis of osteosarcoma cells and its mechanism

Abstract:Objective To investigate the effect and mechanism of parathyroid hormone type 1 receptor (PTHR1) on the inflammation and angiogenesis of osteosarcoma. Methods Osteosarcoma cells MG63 were cultured and divided into negative control group (si-NC) and interference group (si-PTHR1). The expression levels of PTHR1 and AGT mRNA in si-NC group and si-PTHR1 group were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The expression levels of tumor necrosis factor (TNF), interleukin-1β (IL-1β), and interleukin-8 (IL-8) in the cells of the two groups were detected by Western blot. The angiogenesis ability of the cells in the two groups was detected by angiogenesis assay. Results The relative expression levels of PTHR1 and AGT mRNA in MG63 cells in the si-NC group were 0.94 ± 0.08 and 1.12 ± 0.11, respectively, which were higher than those in the si-PTHR1 group at 0.58 ± 0.06 and 0.55 ± 0.04. The difference was statistically significant (t = 6.24, 8.44, all P values <0.01). The expression levels of TNF, IL-1β, and IL-8 in the two groups were 1.18 ± 0.16 and 0.58 ± 0.04, 1.14 ± 0.13 and 0.56 ± 0.05, 1.18 ± 0.08, and 0.52 ± 0.04. The difference was statistically significant (t=6.30, 7.21, 10.84, all P values <0.01). The angiogenesis rate in si-NC group and si-PTHR1 group was 92.90% ± 6.42 %and 57.27 %± 3.10%. The difference was statistically significant (t =8.66, P<0.001). Conclusion sDown-regulation of PTHR1 in osteosarcoma cells can reduce the release of inflammatory factors and angiogenesis, and its mechanism may be related to the down-regulation of AGT.
