Abstract:Objective To investigate the expression of autophagy-related gene Beclinl, microtubule-associated protein 1 light chain3(LC3-Ⅱ) and P62 in ovarian cancer tissues and explore its clinical significance. Methods Fifty cases of epithelia ovarian cancer excision specimens definite by histopathology selected from our department from June 2011 to March 2013 were experimental group, and 20 cases of normal ovarian specimens as the control. Expressions of LC3-Ⅱ, Beclinl and P62 were detected by immunohistochemistry. The correlations of Beclinl, LC3-Ⅱ and P62 expression to the clinicopathologic characteristics of the 50 epithelial ovarian cancer patients were analyzed. Results The positive rates of LC3-Ⅱ and Beclinl were significantly lower in epithelial ovarian cancer than that in normal ovarian tissue (46.00%, 23/50 vs 95.00%, 19/20;42.00%, 21/50 vs 85.00%,17/20), and the positive rates of P62 were significantly higher in epithelial ovarian cancer than that in normal ovarian tissue (72.00%, 36/50 vs 10.00%, 2/20;P<0.05). The expression of Beclinl and LC3-Ⅱ were associated with FIGO stage and histological grade(P<0.05). The expression of P62 was related to FIGO stage(P<0.05). Beclinl expression was positively correlated with the expression of LC3-Ⅱ(P<0.01). The expression of P62 was negatively correlated with Beclinl and LC3-Ⅱ expression(P<0.01). Conclusions The abnormal expression of Beclin 1, LC3- II and P62 in epithelial ovarian carcinoma tissues lead to decrease of autophagosome formation and function of autophagy, and may be associated with ovarian cancer occurrence, development and prognosis.
